Automated and Integrated Analysis Workflow for Adverse Outcome Pathway Identification, Hypothesis Generation and Risk Assessment

Regulations limiting the use of new animal testing in cosmetics has made the existing information more valuable, and at the same time the field of toxicology continues to evolve from a purely descriptive to a highly data-rich science. To structure the data available and make the usage more effective, computational approaches for data integration should be applied. We aim to do this by building upon a previous framework for deriving in vivo toxicity responses from in vitro data and establishing an automated workflow for Adverse Outcome Pathway (AOP) hypothesis generation. In both cases we combine and integrate data coming from diverse sources (pathway data, disease, and in vitro transcriptomics data), and we demonstrate in practice how to use computational approaches for integration of pre-existing knowledge. Upon further refinement, this approach could support chemical risk assessment, identify data gaps where additional testing may be needed, and guide the development of an integrated testing strategy in regulatory settings.
